For some particularly hard or tough samples, blood vessels, etc., it is difficult to use ordinary mechanical grinding methods to ensure the grinding and crushing effect and the samples are always in a low temperature state. Although the traditional manual grinding method can take care of both, when the number of samples is relatively large, manual grinding is very time-consuming and laborious, and there is often the possibility of cross-contamination. If extracting RNA and enzymes that are sensitive to temperature, it is very important to use liquid nitrogen for pretreatment to ensure that the sample is kept at low temperature during the crushing process.

The Department of Vascular Surgery in Changhai Hospital is led by my country’s well-known expert in vascular surgery, Professor Jing Zaiping, and is the top priority of the PLA Institute of Vascular Surgery, a key discipline of the National 211 Project, and Shanghai Clinical Medical Center for Vascular System Diseases.

The discipline is mainly dedicated to the clinical and research work of vascular system diseases, especially in the diagnosis and treatment of vascular diseases such as aortic valve stenosis, aortic dissecting aneurysm, abdominal aortic aneurysm, carotid artery stenosis, lower extremity arterial embolism and other vascular diseases. level.

The center purchased a Tissuelyser-48 automatic sample rapid grinder to facilitate the fragmentation of rat blood vessel tissues and human blood vessel cancerous tissues.

There is no significant difference in the concentration and purity of the DNA and RNA extracted from FT after the automatic sample rapid grinder treatment and manual liquid nitrogen grinding treatment of Shanghai Jingxin (pairwise test P>0.05). However, manual grinding has a larger sample loss rate. For samples with low DNA extraction concentration, the automatic grinder can achieve zero sample loss. It has obvious advantages in DNA extraction, and the manual processing process is complicated and easy to cross. Contamination, operation time and other aspects are not as good as Shanghai Jingxin's automatic sample rapid grinder, so the automatic grinder has great advantages for DNA extraction compared to manual grinding.
